This tutorial follows Part B: Compare results from two tracks.
- Scroll down the Analysis view to the Flexibility-Karplus-Schulz track. Click on the plus sign to reveal two sub-plots called Flexibility Regions and Flexibility Plot.
Note that under the current (default) options, there are 8 boxes in Flexibility Regions compared to 6 for Chou-Fasman “Turn Regions”:
By making the options more stringent, we can find which of these areas are most likely to be flexibility regions.
- Open the options for the Flexibility (Karplus-Schulz) track by going to the Tracks panel and clicking on the words “Flexibility (Karplus-Schulz).” Any editable options are displayed in the Track Options section:
- Change the Flexibility threshold to “1.07.”
Note that the Flexibility Regions graph now corresponds almost exactly with Chou-Fasman Turn Regions selected in Part B. This provides additional confirmation that these regions may correspond to calcium binding regions.
- Return to the default options by clicking the Restore Default Values tool ( ).
- Remove the Flexibility (Karplus-Schulz) track from the Analysis view by removing its checkmark in the Tracks panel.
Proceed to Part D: Recolor tracks.
