This tutorial follows Part B: Compare results from two tracks.

  1. Scroll down the Analysis view to the Flexibility-Karplus-Schulz track. Click on the plus sign to reveal two sub-plots called Flexibility Regions and Flexibility Plot.

Note that under the current (default) options, there are 8 boxes in Flexibility Regions compared to 6 for Chou-Fasman “Turn Regions”:

By making the options more stringent, we can find which of these areas are most likely to be flexibility regions.

  1. Open the options for the Flexibility (Karplus-Schulz) track by going to the Tracks panel and clicking on the words “Flexibility (Karplus-Schulz).” Any editable options are displayed in the Track Options section:

  1. Change the Flexibility threshold to “1.07.”

Note that the Flexibility Regions graph now corresponds almost exactly with Chou-Fasman Turn Regions selected in Part B. This provides additional confirmation that these regions may correspond to calcium binding regions.

  1. Return to the default options by clicking the Restore Default Values tool ( ).
  1. Remove the Flexibility (Karplus-Schulz) track from the Analysis view by removing its checkmark in the Tracks panel.

Proceed to Part D: Recolor tracks.
